Late blight is one of the most damaging diseases of tomato and other crops. It is caused by the oomycete Phytophthora infestans and characterized by rapid epiphytotic development, variable pathogen populations and the ability to overcome monogenic plant resistance. The pathogen is capable of completely destroying tomato plants in a short time at any stage of onthogenesis and can infect all above-ground parts of the plant, causing necrosis and death at last. Two physiological races of Phytophthora infestans have been identified in tomatoes: race To (less aggressive) and race T1 (more aggressive). The most effective method of disease management is the cultivation of resistant tomato varieties and hybrids. Currently molecular genetic marking is being actively used to evaluate plant material for resistance genes, that allow to reduce the use of artificial infection backgrounds. Genetic markers make the opportunity to the identification of resistant genotypes at the early stages of the breeding process and to accelerate the breeding process. The present study is dedicated to molecular genetic evaluation of Solanum lycopersicum breeding samples for homologs of the late blight resistance Ph-3 locus. The results of the study allow to identify resistant variants for further breeding. DNA was analyzed by PCR with three pairs of gene-targeted SCAR primers: SCAR-Ph3-1, SCAR-Ph3-2, and SCAR-Ph3-3. The amplification products were separated by electrophoresis in an agarose gel. The resulting profiles allowed samples to be differentiated by the presence of diagnostic fragments associated with homologs of Ph-3a–Ph-3d. Samples that showed a consistent resistance profile across three primer pairs are recommended for inclusion in further plant breeding process. However genotyping should be supplemented with sequencing of functionally significant regions of Ph-3 and testing against an infectious background, as the presence of a homolog or linked marker does not always equate to the presence of a functional resistance allele.
